Objective    To observe the effect of noggin protein on proliferation and invasion of breast cancer MCF-7 cells.     Methods    MCF-7 cells were infected with recombinant noggin adenovirus. Effect of noggin protein on proliferation of MCF-7 breast cancer cells was detected by MTT assay. Changes in movement and invasion of MCF-7 breast cancer cells were examined by cell scratching test and trans-well cell invasion experiment. Expression of MMP-1 and CXCR4 was assayed by real-time PCR and Western blotting.     Results    The proliferation of MCF-7 cells in over-expressed noggin protein was increased（P0.05）. Scratching test showed that the healing of scratched MCF-7 breast cancer cells was delayed in over-expressed noggin protein. Trans-well cell invasion experiment showed that the number of MCF-7 breast cancer cells that penetrated the cell membrane was significantly less in noggin group than in GFP group (25±3 vs 55±4

P0.05）. Real-time PCR and Western blotting showed that the expression of CXCR4 and MMP1 was down-regulated in MCF-7 breast cancer cells when the noggin protein was over-expressed.     Conclusion    Noggin protein can inhibit the movement and migration of MCF-7 breast cancer cells by down-regulating the expression of CXCR4 and MMP1.
