Objective To observe the expression of NgR in the brain and the neuroethology of rats with experimental autoimmune encephalomyelitis(EAE) after Ad-shRNA-NgR was transfected into the lateral cerebral ventricle of rats. Methods One hundred and eight Wistar rats were randomly divided into normal control group
blank control group
negative control group
and specific RNAi group
27 rats in each group. Rats in blank control group
negative control group
and specific RNAi group were injected with PBS
Ad-shRNA-HK and Ad-shRNA-NgR
respectively
in 7 d after EAE model establishment. Expression of NgR in brain tissue of rats was detected by RT-PCR with immunofluorescence staining after 7 and 14 d
respectively. Neurologic function was scored according to the Koh scoring system. Results The expression of NgR mRNA and the IOD of NgR in specific RNAi group was significantly lower than the blank or negative groups (P0.01) at the 7th day
and was higher in 14 d than in 7 d after injection of different substances (P0.05) and was still lower than in blank or negative group (P0.05) . The latent period of EAE was longer and the neurologic score was lower in specific RNAi group than in blank or negative group(P0.05). No significant change was observed in body weight of rats. Conclusion Adenovirus-mediated NgR specific RNAi can down-regulate the expressing of NgR and ameliorate the neural behaviors of rats with EAE.
