Objective To study the correlation between Runx3 methylation in gastric cancer tissue
H.pylori infection
clinicopathologic features and survival rate of gastric cancer patients. Methods Eighty-six patients with primary gastric cancer (52 males and 34 females) at the age of 28-78 years (mean 52 years)
admitted to Department of Oncology Surgery from June 2004 to December 2005
were selected as an experimental group
and 20 patients admitted to Department of Gastroenterology during the same period were selected as a control group. Runx3 methylation was detected by methylation specific (MSP) PCR in gastric cancer tissue
its adjacent and normal gastric tissues. H.pylori infection was detected by fast urea enzyme test with Warthin-Starry staining in experimental and control groups. Survival rate was calculated by Kaplan-Meier survival curves. Multivariate survival analysis was performed with Cox proportional hazard regression model. Results The Runx3 methylated rate was significantly higher in gastric cancer tissue (65.1%,56/86) than in adjacent gastric tissue (8.1%,7/86) and normal gastric tissue (0/20)(P0.01). The Runx3 methylation was significantly correlated with tissue differentiation
depth of invasion
and lymph node metastasis (P=0.05). The Runx3 methylation rate was higher in gastric cancer tissue infected with H.pylori than in those not infected with H.pylori (P=0.003). Kaplan-Meier survival curves and Log rank test showed that Runx3 methylation was correlated with survival rate of gastric cancer patients (P0.01). Multivariate survival analysis revealed that depth of invasion and Runx3 methylation were the independent prognostic factors for gastric cancer. Conclusion Runx3 methylation is correlated with the development and metastasis of gastric cancer. H.pylori infection increases Runx3 methylation
and Runx3 methylation is an independent prognostic factor for gastric cancer.
