Objective To explore the effect of chemokine receptor CXCR7 and its ligand CXCL12 on the migration
invasion and adhesion of breast cancer cell line MDA-MB-435s. Methods The expression of the CXCR7 on MDA-MB-435s cells was inhibited by RNA interference. Expression levels of CXCR7 mRNA and CXCR7 protein were assayed by RT-PCR and Western blot respectively. Effect of altered expression of CXCR7 on cell migration was measured by cell scratch assay
cell invasion by Transwell chamber and cell adhesion by adhesion assay respectively. Results The expression levels of CXCR7 mRNA and CXCR7 protein
and the migration
invasion and adhesion of the cancer cells were all decreased significantly after the silencing of CXCR7 compared with the control cells. Conclusion The migration
invasion and adhesion of human breast cancer cells are regulated by CXCR7. CXCL12/CXCR7 axis may play an important role in the invasion and metastasis of breast cancer.
