摘要

目的探讨抗肿瘤新药诺帝(Nordy)在体外对卵巢癌细胞(3AO黏液性癌， SKOV3浆液性癌)生长的抑制作用及对细胞周期的影响，同时对卵巢癌细胞株中Aurora-A的mRNA和蛋白过表达的抑制作用进行研究。方法 MTT法检测诺帝对细胞增殖的抑制作用；RT-PCR和Western blot检测Aurora-A的mRNA和蛋白表达情况；流式细胞术分析细胞周期。结果经诺帝作用后Aurora-A的mRNA及蛋白的表达水平显著下降(P0.01)；诺帝能明显抑制3AO、SKOV3细胞的增殖，阻滞细胞周期于G

期（P0.05）。结论诺帝对卵巢癌3AO、SKOV3细胞的生长具有明显抑制作用，可以阻滞细胞周期。在诺帝多环节的抑癌机制中，抑制癌细胞株Aurora-A的异常高表达，阻止中心体的异常转变可能是其机制之一。

Abstract

Objective To investigate the effects of Nordy on the proliferation

cell cycle， and the mRNA and protein expressions of Aurora-A in human ovarian cancer cell lines 3AO and SKOV3. Methods After being treated with Nordy at the doses of 25

phase (P0.05). Conclusion Nordy signi-ficantly inhibits the proliferation and blocks cell cycle of 3AO and SKOV3 cells. One of the underlying mechanisms may be through decreasing the expression of Aurora-A at mRNA and protein levels and preventing abnormal division of centrosome.
