Objective To observe the changes of F-actin of strial capillary endothelial cells from guinea pig cochlea after TNF-α treatment so as to further study the mechanism of permeability of those cells. Methods Strial capillary endothelial cells were dissociated from guinea pig cochlea and cultured respectively with 0.05
0.1
0.2 ng/ml TNF-α for 90 min
then detected by immunofluorescence laser confocal microscopy for F-actin concentrations. The blank control was set simultaneously. Results TNF-α decreased the content of F-actin in strial capillary endothelial cells (P0.01)
and with the increasing TNF-α concentration
the F-actin concentration declined. Conclusion TNF-α may change the density and content of F-actin in strial capillary endothelial cells of guinea pig cochlea.
