Objective To investigate the feasibility of ascorbic acid to induce mouse embryonic stem cells (mESC) to differentiate into cardiac myocytes. Methods Undifferentiated mouse embryonic stem cells grew confluent and were digested by trypsin to single cell to form embryoid bodies (EBs) by rotary cell culture system (RCCS). After plated
EBs were induced by ascorbic acid
then observed with light microscopy and confocal laser scanning microscopy (CLSM)
identified by immunohistochemistry and RT-PCR. Results The cells after induction displayed spontaneous and rhythmic contractile activity. The beating cells was markedly increased in number and accounted for 90% of the induced cells. They were positive for cTnT
α-actinin and actin
and also expressed the cardiac genes α-MHC and β-MHC. Conclusion Ascorbic acid could effectively induce mES cells into cardiac myocytes.
