Objective To explore the expression changes of mitochondrial electron transport chain (ETC) complexes Ⅰ,Ⅱ,Ⅲ and Ⅴ induced by chronic hypoxia. Methods Sixteen adult male Wistar rats were randomly and equally divided into hypoxic group (4 500 m
30 d) and the normoxic control. Hibateral gastrocnemii were collected and mitochondria were isolated. Respiratory state 3 (ST3)
state 4 (ST4) and respiratory control rate (RCR) were measured with Clark oxygen electrode. Mitochondrial ETC complexes Ⅰ,Ⅱ,Ⅲ and Ⅴ expressions were measured with Western blot. Results ST3 and RCR were significantly lower in hypoxic group than in the control. Mitochondrial ETC complexes Ⅰ,Ⅱ,Ⅲ and Ⅴ expressions were significantly lower in hypoxic group than in the control. No significant difference of ST4 and complex Ⅲ core subunit 2 was found between the 2 groups. Conclusion Non-concordant expression of gastrocnemius mitochondrial ETC complexes can be induced by chronic hypoxia
resulting in the decrease of mitochondrial oxidative phosphorylation function.
