摘要

目的采用AdEasy系统构建的带有标记基因RFP的重组腺病毒Ad-Cyr61 感染人肝癌细胞株HepG2

并观察外源性Cyr61 对肿瘤细胞生长和迁移的影响。方法 Ad-Cyr61 与Ad-RFP 分别感染人肝癌细胞株HepG2

荧光倒置显微镜观察荧光表达

通过 RT-PCR、免疫组化实验分别检测Ad-Cyr61 的mRNA和蛋白的表达；通过MTT实验、细胞计数实验、克隆形成实验和划痕愈合实验

观察Cyr61 对人肝癌细胞株HepG2细胞增殖和迁移的影响。结果 Ad-Cyr61 感染人肝癌细胞株HepG2

与对照组比较细胞数量增多

集落形成增加

细胞划痕逐渐愈合。结论 Ad-Cyr61 感染人肝癌细胞株HepG2后可促进细胞增殖和迁移

提示Cyr61 在肿瘤发生和转移中可能起到重要作用。

Abstract

Objective To observe the proliferation and migration of human hapetocelluar carcinoma cell line HepG2 after the introduction of exogenous cysteine rich 61 (Cyr61). Methods Adenviral vector

AdEasy with a report gene

red fluorescence protein (RFP) was used to construct recombinant vector Ad-Cyr61. The vectors Ad-Cyr61 and Ad-RFP (control) were transformed into HepG2 cells to observe the protein and mRNA expression of Cyr61 by immunohistochemistry and RT-PCR. MTT

plate colony formation and cell wound healing assay were performed to determine the Cyr61’s effect on the proliferation and migration of HepG2. Results Ad-Cyr61 transfected HepG2 cells expressed exogenous Cyr61

formed more clones

proliferated more cells and exhibited a greater ability to migrate in cell wound healing than the control cells. Conclusion Exogenous Cyr61 stimulates the proliferation and migration of HCC cells

suggesting that Cyr61 may play a role in tumorigenesis and metastasis.
