摘要

目的研究钙调神经磷酸酶(calcineurin

CaN)与皮层神经元轴突生长的相关性。方法原代培养大鼠大脑皮层神经元。细胞分为正常对照组和环孢霉素A (Cyclosporin A

CsA)处理组。CsA处理组是在细胞更换无血清的NB培养基2 h后加入10 nmol/L CsA刺激。采用倒置相差显微镜和数码影像采集系统观察培养3、7、10 d的神经元，并用SPOT软件测量神经元轴突长度。CaN活性采用酶底物显色的方法进行检测。结果 10 nmol/L CsA刺激后3 d和7 d的神经元轴突均能较显著地增长(P0.05)，而这种作用在10 d更为明显(P0.01)；而所有时相点神经元CaN的活性均受到显著抑制(P0.01)。CsA处理组各时相点CaN活性与皮层神经元轴突的长度均呈显著负相关（r=-0.994

P0.01）。结论 CaN活性与皮层神经元轴突的生长呈负相关。

Abstract

Objective To study the relationship between calcineurin (CaN) activity and axon growth. Methods The cerebral cortical neurons of rats were cultured

and grouped as control group and cyclosporin A (CsA) group in which 10 nmol/L CsA was given when serum-free NB culture media was adopted. Inverted phase contrast microscope and digital photo collection system were used to harvest the photos of neurons cultured for 3

10 d. The length of axons were measured by SPOT software. The CaN activities were measured by enzyme substrate colormetric methods. Results After CsA stimulation

the length of axons increased significantly at 3

7 d (P0.05) and more notably at 10 d (P0.01)

meanwhile the CaN activities were more significantly inhibited at all time points (P0.01). Conclusion CaN activities have a negative relationship with axon growth.
