摘要

目的研究骨形态发生蛋白（BMP）-2和BMP-6对人骨肉瘤细胞株MG63和U2OS的作用。方法分别用重组腺病毒AdBMP-2、AdBMP-6和AdGFP感染骨肉瘤细胞MG63和U2OS，利用台盼蓝拒染法、TUNEL法、吖啶橙/溴乙啶（AO/EB）荧光双染法、Transwell小室和碱性磷酸酶（ALP）活性测定法分别检测细胞增殖、凋亡、迁移以及成骨分化能力的变化。结果与感染AdGFP的相应骨肉瘤细胞株（对照组）相比，分别感染AdBMP-2和AdBMP-6的两种骨肉瘤细胞株的细胞存活率随时间延长逐渐降低，凋亡率逐渐增加，细胞穿膜数逐渐减少，ALP活性逐渐增加。结论 BMP-2和BMP-6可以抑制人骨肉瘤细胞株MG63和U2OS的增殖和迁移，并诱导它们凋亡和向成骨细胞分化。

Abstract

Objective To investigate the effects of BMP-2 and BMP-6 on osteosarcoma cell line MG63 and U2OS. Methods The two osteosarcoma cell lines were infected with AdBMP-2

AdBMP-6 (as experimental groups)

and AdGFP (as control)

respectively. Abilities of their proliferation

transmigration

apoptosis and differentiation were detected by trypan blue exclusion test

transwell-room

acridine orange-ethidium bromide fluorescent staining

in situ cell death detection and the test of alkaline phosphatase activity respectively. Results In the experimental groups

the cell survival rates were significant decreased (P0.01); the apoptosis indexes were significantly increased in a time-dependent manner (P0.01); the cell numbers of transmembrane were decreased (P0.01); and the activity of alkaline phosphatase was increased gradually (P0.01). Conclusion The human BMP-2 and BMP-6 could inhibit the proliferation and migration

promote apoptosis

and reverse the malignancy of osteosarcoma cell line MG63 and U2OS.
