摘要

目的建立高效液相色谱法测定黄连中生物碱含量的方法。方法色谱柱为C

柱(3.9 mm×300 mm，10 μm)，流动相为乙腈：0.05 mol/L磷酸二氢钾（40∶60，内含0.015 mol/L十二烷基磺酸钠），检测波长为345 nm。结果在以上色谱条件下，小檗碱、巴马汀、黄连碱、表小檗碱和药根碱分离效果好。结论方法灵敏度高，重现性好，可用于黄连等含小檗碱型生物碱中药材及制剂的质量控制。

Abstract

Objective To establish an HPLC method for separation and determination of the alkaloids in Rhizoma Coptidis. Methods HPLC system consisted of C

column (3.9 mm×300 mm

mobile phase was composed of acetonitrile solution of potassium phosphate (0.05 mol/L

40∶60

with 0.015 mol/L SDS)

and the detection wavelength was 345 nm. Results Five kinds of berberine-type alkaloids which are jatrorrhizine

epiberberine

coptisine

palmatine and berberine could be separated completely. Conclusion HPLC is of high sensitivity and good reproducibility. It can be used for quality control of drug and Chinese crude drug containing alkaloids.
