摘要

目的观察大鼠血管平滑肌α肌动蛋白基因反义腺病毒载体（pAdanti-α-SMA）转染对转化生长因子β1（TGF-β1）所诱导的肾小管上皮细胞-肌成纤维细胞转分化（TEMT）的影响。方法 pAdanti-α-SMA载体构建并鉴定，转染肾小管上皮细胞（TEC）后免疫荧光染色和Western blot检测α-SMA水平。实验分为5组

A组：正常肾小管上皮细胞（TEC，对照组）；B组：TEC＋2 μl pAdanti-α-SMA；C组：TEC＋1 μg天然TGF-β1；D组：TEC＋1 μg天然TGF-β1＋2 μl pAdanti-α-SMA；E组：TEC＋1 μg天然TGF-β1＋10 μl pAdanti-α-SMA。结果成功构建pAdanti-α-SMA，pAdanti-α-SMA转染TEC后抑制了TGF-β1诱导的TEC 的α-SMA水平并呈剂量依赖性。结论 pAdanti-α-SMA可有效地抑制肾小管上皮细胞的肌成纤维细胞转分化。

Abstract

Objective To evaluate the inhibiting effect of antisense vascular smooth muscle alpha adenovirus vector on tubular epithelial myofibroblast transdifferentiation induced by TGF-β1. Methods The pAdanti-α-SMA was constructed and identified. The α-SMA expression in tubular epithelial cells (TEC) was detected by immunofluorescence and Western blot. Five groups were designed as control (A)

TEC transfected by 2 μl pAdanti-α-SMA (B)

TEC induced by TGF-β1 (C)

TEC induced by TGF-β1 and transfected by 2 μl pAdanti-α-SMA (D)

TEC induced by TGF-β1 and transfected by 10 μl pAdanti-α-SMA (E). Results The pAdanti-α-SMA was successfully constructed. The expression of α-SMA in TEC induced by TGF-β1 was inhibited by pAdanti-α-SMA. Conclusion Tubular epithelial myofibroblast transdifferentiation can be inhibited by pAdanti-α-SMA.
