摘要

目的研究诺帝（Nordy）对HPV16型亚基因(E6、E7)永生化人宫颈上皮细胞（H8细胞）的诱导分化作用。方法 MTT法检测诺帝对H8细胞增殖的抑制作用，流式细胞术（FCM）分析细胞周期，免疫细胞化学S-P法检测增殖细胞核抗原Ki67的表达，TRAP-ELISA法检测端粒酶活性变化。结果诺帝能够明显抑制H8细胞增殖；可阻滞细胞周期于G

期，S期细胞减少；细胞核内Ki67蛋白的表达水平明显下降；端粒酶活性明显降低。结论诺帝对H8细胞有诱导分化作用，这种作用可能是通过抑制增殖，阻滞细胞周期，降低端粒酶活性来实现的。

Abstract

Objective To investigate the effects of Nordy on induced differentiation of HPV16 subgenes-immortalized human endocervical cells (H8 cells). Methods After treated with Nordy

the proliferation of H8 was tested with MTT assay. The effect of Nordy on cell cycle of H8 was analyzed by flow cytometer(FCM). The expression of Ki67 of H8 was detected by immunohistochemistry. The activity of telomerase was tested by TRAP-ELISA. Results Nordy could inhibit proliferation of H8 cells significantly. H8 cells were arrested in G

phase after Nordy treatment. The Ki67 expression and telomerase activity reduced significantly after Nordy treatment. Conclusion Nordy could induce the differentiation of H8 cell lines by inhibiti
