Qin HUANG, Zhenzhen ZHANG, Quanbo LIU. Effect of long intergenic non-coding RNA 238 on HBV replication and expression
Objective
2
To determine the regulating role of long intergenic non-coding RNA 00238 (Linc00238) in the replication of hepatitis B virus (HBV) and explore the potential mechanism.
Methods
2
HBV positive and HBV negative liver tissues were proceeded to LincRNA array
and then the interested Linc00238 was confirmed further by real-time PCR in HBV expression cell line HepAD38
HepG2 cells as well as HBV transient transfection cell line HepG2.2.15. After overexpression and RNA interference techniques
the effect of Linc00238 on the replication
transcription and expression of HBV was determined by gain-of-function and loss-of-function study in HepAD38 cells. Dual-luciferase reporter assay was used to detect the effect of Linc00238 on promoter activity of HBV.
Results
2
LincRNA array indicated there were 13 LincRNAs having changed expression levels in HBV positive liver tissues
with 2 up-regulated and 11 down-regulated. Real-time PCR further showed the expression level of Linc00238 was significantly down-regulated in HBV positive liver tissues
HBV expression cell line and HBV transient expression HepG2.2.15 cells (
P
<
0.05)
which indicating that HBV infection inhibited the expression of Linc00238. Overexpression of Linc00238 suppressed HBV replication and protein levels significantly in HepG2 (
P
<
0.05)
and while sh00238 inte
rference restored the HBV replication and protein levels (
P
>
0.05). The reporter luciferase reporter system showed no changes were found after the co-transfection of pHBs-luc
pHBx-luc
pHBc-luc and pCDNA-linc00238
indicating that Linc00238 has no effect on HBV promoter activity (
P
>
0.05).
Conclusion
2
Linc00238 exerts inhibitory effect on HBV protein and DNA expression
but this effect is not associated with HBV promoter activities.
