Expression, clinical significance and functional roles of LHX3 gene in hepatocellular carcinoma
clinical significant and functional roles of LIM homeobox 3 (LHX3) in hepatocellular carcinoma (HCC).
Methods
2
Atotal of 100 HCC tissue samples from the patients with fully clinicopathological data and 45 paired para-cancerous tissues were collected for tissue microarray. PCR
Western blotting and immunohistochemical staining were used to detect the expression of LHX3 in the HCC tissues and para-cancerous tissues. The differences of LHX3 expression in different groups and the relationship of the expression with clinicopathological data were analyzed. Kaplan-Meier and Cox regression survival analyses were performed to investigate the prognostic significance of the gene in total survival time. Transwell assay was employed to study the functional roles of LHX3 in tumor metastasis.
Results
2
LHX3 was highly expressed in the HCC tissues compared to normal or para-cancerous tissues. The expression was significantly correlated with clinical stage (
P
=0.037) and metastasis (
P
<
0.001)
but not with age
gender
histological grade
tumor size and tumor location (
P
>
0.05) of HCC patients. Its expression was remarkably increased in the HCC patients at advanced stages or with metastasis. Further survival analyses showed that the increased LHX3 expression level was associated with unfavorable prognosis (
P
=0.002)
and could be regarded as an independent prognostic factor for HCC patients (haz
ard ratio=1.265
P
=0.032). Functional studies indicated that LHX3 over-expression promoted cancer cell migration and invasion in HCC cells.
Conclusion
2
LHX3 shortens the total survival time of HCC patients through promoting metastasis.
