Electrospinning membranes with different microscopic morphologies: hydrophilicity, degradation, and influence on osteoblast proliferation and differentiation
To prepare electrospinning nanofiber membranes with different microscopic morphologies and compare their hydrophilicity
degradation
and influences on the proliferation and differentiation of osteoblasts.
Methods
2
Different electrospinning nanofiber membranes were prepared by adjusting the parameters
and the microscopic morphologies of the membranes were observed using scanning electron microscope and inverted phase-contrast microscope. The hydrophilicity and degradation of these membranes and their influences on the proliferation and differentiation of osteoblasts were analyzed using a visual contact angle test system
weight loss test
cell proliferation assay
ALP activity assay and qRT-PCR.
Results
2
Under scanning electron microscope (×500) and inverted phase-contrast microscope (×100)
the electrospinning nanofiber membranes prepared with 15%
10%
and 8% PCL showed a filamentous morphology
containing a small amount of beads
and a predominantly bead-like morphology
respectively. The 3 membranes exhibited similar hydrophilicity with also similar influences on osteoblast proliferation. The bead-like membrane had the highest degradation rate
followed by the bead-containing filamentous membrane and then by the filamentous membrane. The degree of osteoblasts differentiation on the 3 membranes was lowered in the order filamentous membrane
bead-containing filamentous membrane and bead-like membrane.
Conclusion
2
Electrospinning membrane with different microscopic morphologies can be prepared by adjusting the electrospinning parameters. Compared with the bead-containing and bead-like electrospinning membranes
the filamentous membrane is characterized by a slower degradation rate and a better capacity to promote the differentiation of osteoblasts.
