Effect of miR-455-3p overexpression on migration and invasion of colon cancer HCT116 cells and possible mechanism
To investigate the effect of miR-455-3p overexpression on the migration and invasion of colon cancer HCT116 cells and explore the underlying mechanism.
Methods
2
HCT116 cells were transfected with miR-455-3p mimics or miR-455-3p NC using Lipofectamine
TM
3000
and the expression of miR-455-3p in t
he cells was detected with real-time PCR. The changes in the viability of the transfected cells was assessed using MTT assay
and the alterations in the cell migration and invasion abilities of the cells were investigated using scratch wound healing assay and Transwell assay
respectively. The expression levels of matrix metalloproteinases 2 (MMP2)
MMP9
and the proteins related with the phosphatidylinositol 3 kinase (PI3K)/protein kinase B (AKT) signaling pathway were detected using Western blotting.
Results
2
Compared with miR-455-3p NC
transfection with miR-455-3p mimics resulted in significantly up-regulated expression of miR-455-3p (
P
<
0.01)
obviously lowered cell viability and migration and invasion abilities (
P
<
0.01)
and significant down-regulation of the expression of MMP2
MMP9
PI3K and p-AKT (
P
<
0.01) in HCT116 cells.
Conclusion
2
Overexpression of miR-455-3p can inhibit the migration and invasion of HCT116 cells
in vitro
possibly by blocking the PI3K/AKT signaling pathway.
