Expression of circRNA-IARS in pancreatic carcinoma and its clinical significance
To investigate the expression of circRNA-IARS in pancreatic carcinoma and its relation with the clinicopathological features and prognosis of the patients.
Methods
2
The differential expressions of circular RNA (circRNA) between pancreatic cancer cell lines Hs 766T and Hs 766T-L2 were analyzed using gene chips
and the microarray results were verified with real-time quantitative PCR (qRT-PCR). Based on these results and using bioinformatic analysis
circRNA-IARS was identified as the potential target circRNA in close relation with the metastasis and invasion of pancreatic carcinoma. The expression levels of circRNA-IARS and linar-IARS were detected in 25 pairs of pancreatic carcinoma tissues and adjacent normal tissues using qRT-PCR. The expression of circRNA-IARS was also detected in 115 pancreatic tumor tissues to analyze its relations with the clinicopathological characteristics and prognosis of the patients.
Results
2
The microarray results showed that 28 circRNA were upregulated in Hs 766T-L2 cells. In the 25 pairs of pancreatic carcinoma and adjacent tissues
the expression of circRNA-IARS was significantly increased in the tumor tissues as compared with the adjacent tissues (
P
<
0.05). In patients with pancreatic carcinoma
a high expression of circRNA-IARS in the tumor was found to closely correlate with lymph node involvement
vascular invasion
liver metastasis and TNM stages (
P
<
0.05)
and the patients with a high expression level of circRNA-IARS had a significantly shorter postoperative survival than those with a low circRNA-IARS expression (
P
<
0.05).
Conclusion
2
CircRNA-IARS is over-expressed in pancreatic carcinoma
and its expression level is closely related to lymph node invasion
vascular invasion
liver metastasis and TNM stage
and affects the prognosis of the patients.
