摘要

目的

分析贵阳市多家医院临床分离的大肠埃希氏菌中产CTX-M-9型产超广谱β-内酰胺酶(extended spectrum β-lactamase，ESBLs)的基因分型。

方法

收集贵阳市多家医院临床分离的大肠埃希菌74株，经确证非重复产ESBLs大肠埃希菌47株，将已扩增的PCR产物进行基因测序并用NCBI数据库中的BLASTN比对测序结果，从而进行基因分型。

结果

74株大肠埃希菌中检出47株产超广谱β-内酰胺酶。PCR扩增结果显示47株ESBLs基因阳性菌株中，33株携带CTX-M-9组基因。随机选取部分PCR扩增阳性产物进行DNA测序分析

结果显示含CTX-M-9组基因阳性菌株的亚型为CTX-M-14型。

结论

CTX-M-9型耐药基因为本研究产ESBLs大肠埃希菌主要基因，CTX-M-14型为其变异亚型。

Abstract

Objective

To analyze the genotypes of CTX-M-9 producing extended spectrum β-lactamase (ESBLs)in

Escherichia coli

isolated from several hospitals in Guiyang City.

Methods

Among the 74 strains of isolated form several hospitals of Guiyang City

47 strains were confirmed as non repetitive ESBLs producing. Then the amplified PCR products were sequenced

and the results were compared with the BLASTN sequencing results in the NCBI database for genotyping.

Results

In the 74 isolated strains of

Escherichia coli

strains were confirmed to produce ESBLs. PCR amplification showed that 33 of the 47 ESBLs positive strains carried CTX-M-9 group genes. For the randomly selected PCR positive samples

DNA sequencing results showed that the subtype of CTX-M-9 positive strains was CTX-M-14.

Conclusion

Drug resistance gene CTX-M-9 is the major gene in ESBLs producing

Escherichia coli

and CTX-M-14 is its variant subtype.
