Long-chain non-coding ATB regulates invasion and metastasis of breast cancer cells by inhibiting miR-200c expression
To determine the effect of long-chain non-coding ATB on the invasion and metastasis of breast cancer cells
and investigate its underlying mechanism.
Methods
2
The expression levels of ATB and miR-200c in different breast cancer cell lines (BT474
SKBr-3
MCF7 and MDA-MB-453) were detected by qPCR. Double luciferase reporter gene was used to detect the interaction between the 2 molecules. MTT assay and Transwell invasion assay were used to detect the proliferation and invasion of breast cancer cells after ATB inhibition. The reversing effect of miR-200c on the proliferation and invasion of breast cancer cells after ATB inhibition was also detected. Western blotting was used to detect the expression of ZEB1 and ZNF217 protein after ATB inhibition.
Results
2
Compared with other breast cancer cells
SKBr-3 cells had lowest ATB and highest miR-200c expression levels. ATB could bind specifically to the site of miR-200c and regulate its expression activity. Inhibition of ATB enhanced the proliferation and invasion of breast cancer cells
but silencing miR-200c could reverse the effects of ATB inhibition to a certain extent. After ATB inhibition
the protein expression levels of ZEB1 and ZNF217 were restored partially.
Conclusion
2
ATB plays an important role in the incidence and development of breast cancer. ATB targets miR-200c and then regulates the proliferation and invasion of breast cancer cells.
