Correlation of circular RNA hsa_circ_0029642 with pulmonary hypertension in adult patients with congenital heart disease
To investigate the correlation between serum level of circular RNA hsa_circ_0029642 and pulmonary hypertension in adult patients with congenital heart disease and explore the possibility of using hsa_circ_0029642 as a serum marker of pulmonary hypertension.
Methods
2
Serum samples were collected from 20 patients with congenital heart disease and concurrent pulmonary hypertension (group A)
20 patients with congenital heart disease but without pulmonary hypertension (group B)
and 20 patients with rheumatic heart disease and pulmonary hypertension (group C). The total RNA was extracted from the serum samples for reverse transcription to obtain the corresponding cDNA. Real-time fluorescent quantitative PCR reaction was performed to detect the levels of circular RNA hsa_circ_0029642 with GAPDH as the internal control. The RQ value was calculated and the ROC curve was mapped based on the results of quantitative PCR to assess the value of hsa_circ_0029642 in the diagnosis of pulmonary arterial hypertension.
Results
2
The total RNA extracted from the serum samples all had good quality in the 3 groups. The RQ value differed significantly between groups A and B (
P
<
0.05) and between groups B and group C (
P
<
0.01)
but not between groups A and C (
P
>
0.05); the RQ value was significantly lower in groups A and C than in group B
and was comparable between groups A and C. Analysis of the ROC curve showed that the serum level of hsa_circ_0029642 had an area under the curve (AUC) of 0.752 (95%
CI
:0.624~0.884
P
<
0.01) for the diagnosis of pulmonary arterial hypertension.
Conclusion
2
The serum level of circular RNA hsa_circ_0029642 is obviously lowered in patients with congenital heart disease and concurrent pulmonary arterial hypertension and can be used as a new serum marker for pulmonary hypertension.
