Objective
2
To investigate the role of Toll-like receptor 4 (TLR4)/nuclear factor-κB (NF-κB) signaling pathway in osteoblast-like differentiation and calcification of vascular smooth muscle cells (VSMCs) induced by high phosphate.
Methods
2
Cultured mouse VSMCs were exposed to high phosphorus and calcium deposition in the cells was observed with Alizarin red S staining. The effect of pyrrolidine dithiocarbamate (PDTC
20 μmol/L)
a NF-κB inhibitor
on osteogenic differentiation and calcification of the VSMCs with or without exposure to high phosphorus was assessed. The cells were transfected with a small interfering RNA (siRNA) targeting TLR4 or a control siRNA to test the effects of TLR4 knockdown on osteogenic differentiation and calcification of VSMCs exposed to high phosphorus. Western blotting was used to detect the expression of TLR4
p-NF-κB
SM22α
bone morphogenic protein 2 (BMP2) and runt-related transcription factor 2 (Runx2) in the cells with different treatments.
Results
2
Compared with the normal control cells
the VSMCs exposed to high phosphorus exhibited significantly increased calcium deposition (0.079±0.011
vs
0.193±0.017
P
<
0.01) and expression levels of TLR4 (0.228±0.008
vs
0.526±0.017
P
<

p-NF-κB (0.271±0.003
vs
0.454±0.008
P
<
0.01)
BMP2 and Runx2 but with a decreased expression of SM22α. Treatment of the high phosphorus-exposed cells with PDTC significantly decreased calcium deposition in the cells (0.121±0.013
vs
0.217±0.031
P
<
0.01)
lowered the expression of p-NF-κB (0.094±0.006
vs
0.380±0.009
P
<
0.01)
BMP2 and Runx2
and up-regulated the expression of SM22α. Transfection of the high phosphorus-exposed VSMCs with the TLR4 siRNA significantly suppressed the expression of TLR4 (0.117±0.047
vs
0.377±0.012
P
<
0.01)
decreased the expression of p-NF-κB (0.264±0.013
vs
0.601±0.027
P
<
0.01)
BMP2 (
P
<
0.01) and Runx2 (
P
<
0.01)
and increased the expression of SM22α (
P

0.01).
Conclusion
2
High phosphorus can induce osteoblast differentiation and calcification in mouse VSMCs
in vitro
by activating the TLR4/NF-κB signaling pathway.
