Effect of nuclear factor-κB inhibitor on expression of inflammatory factors in lipopolysaccharide-stimulated degenerative human nucleus pulposus cells
摘要
Abstract
Objective
2
To investigate the changes in the expression of nuclear factor-κB (NF-κB) and inflammatory cytokines in degenerative human intervertebral disc nucleus pulposus cells in response to lipopolysaccharide (LPS) stimulation.
Methods
2
Cultured degenerative human nucleus pulposus cells were treated with PBS (control)
LPS (500 μg/mL)
or a NF-κB inhibitor (BAY11-7082) plus LPS (500 μg/mL). The levels of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) in the culture medium were detected with enzyme-linked immunosorbent assay (ELISA). The expression and localization of p65 in the nucleus pulposus cells were detected with immunofluorescence assay
and the cellular expression of TNF-α
IL-1β
NF-κB-binding protein p65 and phosphorylated p65 (p-p65) were detected with Western blotting; Real-time PCR was used to detect the expression of TNF-α and IL-1β mRNA in the cells.
Results
2
LPS stimulation significantly increased the nuclear translocation of p65 and enhanced p-p65 expression in cultured nucleus pulposus cells. LPS stimulation of the nucleus pulposus cells caused sig
nificant increases in the levels of TNF-α and IL-1β both in the culture medium (
P
<
0.05) and in the cells (
P
<
0.05). Compared with LPS-exposed cells
the cells treated with BAY11-7082 plus LPS showed a significant decrease in p65 translocation and p-p65 expression with also decreased levels of TNF-α and IL-1β in both the culture medium (
P
<
0.05) and in the cells (
P
<
0.05).
Conclusion
2
LPS stimulation can promote the expression of TNF-α and IL-1β by activating NF-κB signaling pathway in human degenerative nucleus pulposus cells
and inhibition of NF-κB signaling pathway can significantly reduce the expression of these inflammatory factors.
