Effect of periostin on proliferation of keloid mesenchymal stem cells in vitro
摘要
Abstract
Objective
2
To determine the effect of periostin (also know as osteoblast-specific factor 2) on the proliferation of keloid mesenchymal stem cells (KMLSCs)
in vitro
.
Methods
2
KMLSCs and skin precursors (SKPs) were isolated from keloid tissues and the corresponding skin tissues respectively
and then cultured. The third passage cells were sorted by flow cytometry and then expanded. The expression of periostin was detected by immunocytochemical assay and Western blotting. The shPeriostin lentiviral vector was constructed
and then infected into KMLSCs. The interference efficiency was determined by qPCR and Western blotting. Cell proliferation
apoptosis and cell cycle in the KMLSCs after shPeriostin or blank vector infection and untreated cells were detected by MTT assay and flow cytometry. The expression of LRP5 was determined by immunofluorescent staining and Western blotting.
Results
2
The KMLSCs and SKPs were isolated from keloid and corresponding skin tissues
with
a
proportion of 94%. Immunocytochemical assay and Western blot analysis showed that the expression level of periostin was significantly higher in KMLSCs than in SKPs (
P
<
0.05). qPCR and Western blotting indicated that the expression of periostin was inhibited after the infection of shPeriostin lentiviral vector. Knockdown of periostin resulted in inhibitory proliferation
enhanced apoptosis and more cells arrested at G
1
phase in KMLSCs. Knockdown of periostin also led to the decreased expression of LRP5 (
P
<
0.05).
Conclusion
2
Knockdown of periostin induces apoptosis
inhibits cell cycle transition
and decreases proliferation in KMLSCs
which might be due to its effect on the expression of LRP5.
