Expression of rap1b in hoxb4 transgenic zebrafish during embryo hematopoietic development
摘要
Abstract
Objective
2
To investigate the expression pattern of
rap1b
gene in the zebrafish with
hoxb4
transgene and the relationship between the 2 genes in early hematopoietic development so as to find critical regulatory factors in the early hematopoietic development.
Methods
2
Zebrafish with highly expressed
hoxb4
were selected as the research object. There were 3 groups of zebrafish
namely the exprimental group (zebrafish expressed
hoxb4
-
EGFP
)
the control group (zebrafish only expressed
EGFP
) and the blank control group (wild type zebrafish). Embryo cells with
EGFP
fluorescence signals were sorted on 18 hours post fertilization (hpf)
and 24
30
36 and 48 hpf respectively from control groups and experimental group. The expression of
rap1b
was examined with quantitative real-time PCR (qRT-PCR). Then
the multi-phase embryos of wild type zebrafish were collected to prepare
rap1b
gene antisense mRNA probes
and the hybridization signals of
rap1b
in the 3 gro
ups of zebrafish embryos were observed on 18
24
30
36 and 48 hpf with whole-mount
in situ
hybridization (WISH).
Results
2
The results of qRT-PCR showed that the expression of
rap1b
gene was significantly enhanced on 30
36 and 48 hpf in the experimental group (
P
<
0.05). Antisense mRNA probes of
rap1b
gene were successfully achieved
and WISH results showed that
rap1b
gene was expressed in the range of 18 to 48 hpf in the nervous system and the hematologic system in 3 groups of zebrafish embryos.
Conclusion
2
The expression of
rap1b
gene is obviously increased in
hoxb4
transgenic zebrafish
suggesting that
rap1b
and
hoxb4
may be both involved in the regulation of early hematopoietic development.
