Objective
2
To determine the effect of the cytoplasm retinoic acid receptor α (RARα) expressed in hippocampal cells on learning-memory function in rats.
Methods
2
Sprague-Dawley rats were randomly divided into the Morris water maze (MWM) trained group (
n
=6) and the untrained group (
n
=6). The expression levels of RARα and GluR1 in the hippocampus of the 2 groups were measured after training by PCR and Western blotting. After the RARα shRNAs adeno-associated virus (AAV) and AAV-control shRNA were respectively injected stereotactically into the hippocampus of rats
the learning-memory function was tested by MWM test and the expression levels of RARα and GluR1 in the hippocampus were measured by PCR and Western blotting. After the PC12 cells induced by neuron growth factor (NGF)
in vitro
for 7 d to differentiate into neuron-like cells
the expression levels of RARα and GluR1
the calcium activity and the resting membrane potential level were detected.
Results
2
MWM training enhanced the learning-memory function
and the total protein and cytoplasm protein levels of RARα and GluR1 in the hippocampus when compared with the untrained rats (
P
<
0.01). After the intra-hippocampal injection of AAV-RARα shRNAs for 2 weeks
MWM test results showed that their learning-memory function was statistically decreased than the AAV-control shRNA rats (
P
<
0.05). Meanwhile
the total protein and cytoplasm protein levels of RARα and
GluR1 in the hippocampus were markedly decreased in the AAV-RARα shRNAs injected rats (
P
<
0.01). As a neuron model
in vitro
the resting membrane potential level of PC12 cells induced by NGF treatment was much closer to that of the neurons (
P
<
0.01)
the total and cytoplasm levels of RARα and GluR1 were also obviously increased compared with those of the control cells (
P
<
0.01)
and the cellular calcium activity was enhanced (
P
<
0.05).
Conclusion
2
The cytoplasmic expression level of RARα in the hippocampal cells is positively correlated with the learning-memory ability
and play an important role in regulating synaptic plasticity.
