Objective
2
To establish a human primary dendritic cells(DCs) model infected with dengue virus (DENV).
Method
2
Peripheral blood samples defined with DC genotype of CD209
-139A/-336A were collected. The DC precursor cells were isolated by ficoll-gastrografin density gradient centrifugation and further induced to differentiate into immature DCs by culturing in RPMI1640 media supplemented with recombinant human granulocyte macrophage-colony stimulating factor (rhGM-CSF) and interleukin-4 (rhIL-4). The differentiation efficiency was determined by flow cytometry. The morphologic and biological characteristics of DCs were identified by Wright’s staining and lymphocyte proliferation assay
respectively. Finally
DENV-1 strain GZ2002 was used for construction and evaluation of the DENV infection in human DCs.
Results
2
The immature DCs with typical morphologic characteristics were successfully obtained from the isolated precursor cells with cytokine stimulation
of which the differentiation efficiency was greater than 90%. The acquired DCs could effectively stimulate lymphocyte proliferation and support the entire viral lifecycle during DENV infection
suggesting the successful establishment of DENV infected DC model.
Conclusion
2
DENV infection model based on human primary DCs is successfully established.
