摘要

目的　研究annexinⅡ基因反义表达载体 (annexinⅡ pLXSN)对人肺腺癌细胞株SPC A 1生长的影响。方法　①提取SPC A 1细胞总RNA

应用RT PCR法扩增目的片断

通过双酶切定向克隆的方法构建annexinⅡ基因反义表达载体。②应用脂质体将annexinⅡ pLXSN导入SPC A 1细胞 (SPC A 1 annexinⅡ )。半定量RT PCR法分析转染细胞中annexinⅡmRNA的表达。③观察反义RNA对SPC A 1细胞生物学行为的影响。结果　①反义表达载体成功导入SPC A 1细胞 ;半定量RT PCR证实SPC A 1 annexinⅡ细胞中annexinⅡmRNA表达量较亲代细胞下降约 2 /3。②对转染前后的细胞观察表明

SPC A 1 annexinⅡ细胞生长减慢

克隆形成率降低

3 H TdR掺入率下降

细胞增殖被阻滞于G0 ～G1 期。结论　annexinⅡ具有促进肺癌细胞增殖的作用

此功能将有助于肺癌的发生发展
