Activation of glial cell pyroptosis signal in children with sepsis and its influence on mitochondrial injury in neural cells
To investigate the activation of glial cell pyroptosis signal in children with sepsis-related encephalopathy (SAE) and its influence on mitochondrial injury in neural cells.
Methods
2
A total of 26 patients with sepsis who were admitted to
the First Affiliated Hospital of Hebei North University from April 2020 to March 2024 were enrolled, among whom there were 15 SAE patients and 11 non-SAE patients. Quantitative PCR was used to measure mitochondrial DNA(mtDNA) in serum, and ELISA was used to measure the serum levels of S100 calcium-binding protein B(S100B) and neuron-specific enolase(NSE). BV2 cells were divided into Con group,mtDNA group,si-cGAS group,mtDNA+si-cGAS group,mtDNA+cGAS-oe group, and mtDNA+cGAS-oe+YVAD-cmk group. Western blotting was used to analyze nuclear factor kappa-B/NOD-like receptor pyrin domain containing 3(NF-κB/NLRP3) signal, and flow cytometry was used to measure cell pyroptosis.
Results
2
The SAE group had a significantly higher serum level of mtDNA than the non-SAE group in days 4 and 7 of hospitalization(
P
<
0.05). The serum level of mtDNA was positively correlated with the serum levels of NSE and S100B(
P
<
0.05). Transfection of si-cGAS significantly reversed the effect of mtDNA on NF-κB/NLRP3 signal activation and cell pyroptosis in BV2 cells(
P
<
0.05). Compared with the mtDNA group, the mtDNA+cGAS-oe group had significant increases in the protein expression levels of cGAS,p-NF-κB,NLRP3,and cleaved caspase-1 and cell pyroptosis rate in BV cells(
P
<
0.05). Compared with the mtDNA+cGAS-oe group, the mtDNA+cGAS-oe+YVAD-cmk group had significan
t reductions in the protein expression level of cleaved caspase-1 and cell pyroptosis rate in BV2 cells(
P
<
0.05).
Conclusion
2
The serum level of mtDNA in children with sepsis is closely associated with the changes in the serum levels of NSE and S100B,and the increase in mtDNA may promote the activation of NF-κB/NLRP3 signal and cell apoptosis in BV2 cells in a cGAS-dependent manner.
