Effect of LPS on Akt expression and phosphorylation in mouse lung tissue and RAW264.7 cells
Objective To explore the effect of LPS on the expression and phosphorylation of Akt
a PTEN/PI3k signal transduction downstream molecule
in vivo
and
in vitro
. Methods RAW264.7 cells were attacked with LPS at different concentrations. The Akt protein expression and phosphorylation level were detected by Western blotting at 8
24
48 h after attack. Six h after receiving 20 mg/kg LPS intra-peritoneal injection
the mice were killed and their lung tissues were resected for Akt protein expression and phosphorylation. Results The Akt protein expression of RAW264.7 cells showed no obvious difference after LPS attack at different doses
but their phosphorylation l
evel was increased obviously with the increase of the doses. Whereas the phosphorylation level of Akt protein was decreased in the lung tissue of mice 6 h after LPS attack. Conclusion The phosphorylation level of Akt protein is different after LPS attack
in vitro
and
in vivo
. The Akt protein phosphorylation is increased obviously
