Effects of glucose on phospholipase C and [Ca2+]i in βHC9 cells
Objective To observe the effects of glucose on phospholipase C (PLC) activity and [Ca
2+
]
i
in B cell line βHC9 and determine the role of PLC in glucose-stimulated insulin secretion(GSIS). Methods βHC9 cells were pretreated with various concentrations of glucose
then collected for assay of PLC activity and [Ca
2+
]
i
by using[
3
H]-IP and Fura-2/AM methods respectively. Results When glucose concentration was 1
5
10
15
20 mmol/L
the PLC activity were 8.1、11.0、16.7、19.8、26.4 Bq/ml (
P
<
0.05)
and the[Ca
2+
]
i
were 82
95
128
150 and 161 nmol/L [no significant differences was seen in group 1 and 2
and group 4 and 5
while there were significant differences in the other groups (
P
<
0.05)
]
. Correlation was found between the PLC activity and the [Ca

]
i
(
P

in βHC9 cells
and PLC and Ca
2+
may mediate signal transduction pathway of GSIS in B cells.
