Application of 1.5T magnetic resonance imaging in monitoring glioma orthotopic implantation model in nude mice
Objective To establish a model of human glioma orthotopic implantation in nude mice
and investigate the accuracy and feasibility of 1.5T magnetic resonance in the detection of the transplanted tumor. Methods Glioma cell line SHG-44 was cultured
in vitro
and then the cells (1×10
6
per animal) were injected into the brain parenchyma of 15 nude mice using micro-syringe to form glio
ma. Fifteen days later
these nude mice were scanned by 1.5T MR with animal-coil to detect the tumor. Tumor volume was measured in AW4.3 imaging workstation. Then brain tissue was taken when the mice were dead for HE staining and immunohistochemical staining of CD34. Tumor volume was measured with serial sections under microscope and the results were evaluated with the data of MR imaging. Results Two nude mice were dead in perioperative period
and the rest 13 mice were survived for 20 to 29 d
and MR scanning with contrast injection showed brain tumor were observed in the bran of these mice. The location in MRI was consistent with that in tissue section. Tumor volume was (29.41±10.95) mm
3
measured by MRI and was (26.57±9.70) mm
3
measured under microscope. There was no significant difference (
P
>0.05) but significant correlation (
r
=0.985
P
<
0.01). HE staining and CD34 immunohistochemical staining showed the observed tumor had typical features of glioma. Enhanced imaging displayed better MR images. Diffusion tensor imaging (DTI) showed the distribution of cerebral white matter. Conclusion It is easy and convenient to use micro-syringe directly to establish the human glioma orthotopic implantation model in nude mice. 1.5T MR can detect the tumor in mouse brain in vivo
and carry out certain functional imaging. 1.5T MR is an effective way to dynamically monitor the occurrence and development of glioma in nude mice.
