Pharmacokinetics and relative bioavailability of pyridostigmine bromide sustained-release tablets in rabbits
Objective To study the pharmacokinetics and relative bioavailability of pyridostigmine bromide sustained-release tablets and conventional tablets in rabbits. Methods Six New Zealand rabbits were randomly and evenly divided into two groups. Based on the self-crossover control experiment
the rabbits were subjected to single-dose oral administration of pyridostigmine bromide sustained-release tablets and conventional tablets. The pyridostigmine bromide concentrations in the plasma of rabbits were measured by high-performance liquid chromatography
based on which the pharmacokinetic parameters and relative bioavailability were calculated. Results
After delivery
pyridostigmine bromide sustained-release tablets and conventional tablets had the following pharmacokinetic parameters: t
max
= 2 h and 4 h; C
max
= 17.944 0.991 mg/L and 15.442±0.215 mg/L; AUC
0-∞
= 201.956±4.623 mg/(h·L) and 200.557±3.357 mg/(h·L). When compared with conventional tablets
sustained-release tablets had relative bioavailability of 102.89%. Conclusion Pyridostigmine bromide sustained-release tablets have an obvious sustained-release effect
