Relationship between caveolin-1 and proliferation of basilar artery smooth muscle in rabbit model of subarachnoid hemorrhage
Objective To investigate the expression changes of caveolin-1 and platelet-derived growth factor (PDGF) in the basilar a
rtery of the rabbit model of subarachnoid hemorrhage (SAH) over time and to analyze the relationship between caveolin-1 expression and the proliferation of vascular smooth muscle cells (VSMCs) in basilar artery. Methods Seventy-six New Zealand rabbits were randomly divided into a normal group
a sham group
and SAH groups (
n
=5). Double injection of autologous arterial blood into the cisterna magna was employed to simulate cerebral vasospasm (CVS) in the SAH groups. Sterile saline was used in place of the autologous arterial blood in the sham group. Basilar artery specimens were collected on the 1st
3rd
5th
7th
and 14th days after double injection. Then
the tissues of the specimens were observed using pathological sections
and the mRNA and protein expressions of caveolin-1 and PDGF were investigated through RT-PCR and Western blotting. Results The thickness of vascular smooth muscle layer of basilar artery in the SAH groups increased gradually and peaked on the 7th day
significantly increased as compared with that in the sham group [(39.92±4.07)μm vs (10.25±1.83)μm

and then decreased to a normal value on the 14th day. The mRNA expression of caveolin-1 in the SAH groups peaked on the 5th day
significantly increased as compared with that in the sham group [(63.41±4.47)% vs (11.75±1.84)%
P

. The protein expression of caveolin-1 in the SAH groups decreased to the lowest point on the 7th day
significantly decreased as compared with that in the sham group [(13.78±0.59)% vs (37.63±0.85)%
P

. The mRNA expression of PDGF in the SAH groups peaked on the 3rd day
significantly increased as compared with that in the sham group [(64.45±4.00)% vs (11.13±1.20)%
P

]
. The protein expression of PDGF in the SAH groups peaked on the 7th day
significantly increased as compared with that in the sham group [(40.67±0.81)% vs (15.42±1.00)%

. The protein expression of caveolin-1 was significantly and negative
ly correlated with the thickness of vascular smooth muscle layer of basilar artery (
r

P
