Effect of different La contents in hydroxyapatite-coated Ti plate on cytocompatibility of attached cells
Objective To determine the effect of different La contents in the surface of lanthanum-doped hydroxyapatite (HA)-coated Titanium (Ti) plate on the cytocompatibility of attached cells. Methods Pure Ti plates were coated by means of microarc oxidation in the solutions containing 0
0.3 and 0.8 g/L La
and thus HA coated Ti plate surface with different La contents were obtained. Specimens were seeded with bovine serum albumin. The initial adhesion of protein on the surface was tested by bicinchoninic acid (BCA) protein analyzing. Ti plates with different surface coatings were also seeded with osteoblasts (OB) and cultured in the osteogenic medium. The proliferation and differentiation of OB were evaluated by MTT assay
alkaline phosphatase activity analysis
and scanning electronic microscopy (SEM). Results Ti specimens in 0 g/L La and 0.3 g/L La groups accelerated the early adhesion and spreading of OB on the Ti surfaces. SEM showed that the cells grew well with the Ti specimens and compacted tightly for the groups oxidized in 0 g/L La and 0.3 g/L La contents. The Ti surfaces of these two groups promoted the cells proliferation
and cells grew in a multilayer manner. The modified Ti surfaces enhanced the ALP activity especially on the specimens oxidized in solution with 0 g/L La and 0.3 g/L La contents. The results indicated that the lanthanum doped HA coatings which were oxidized in solutions with La contents of 0 g/L and 0.3 g/L had better compatibility than that oxidized in solutions with La content of 0.8 g/L. Ti oxidized in solutions with 0.3 g/L La and without La improved the adhesion
spreading
proliferation and differentiation of OB on the Ti surfaces. Conclusion Lanthanum-doped HA coating of pure Ti by microarc oxidation improves bioactivity of Ti surface.
