Establishment of human hepatocellular carcinoma cell line stably over-expressing Mov10 and its biological behaviors
Objective To establish a cell line stably expressing Moloney leukemia virus 10 (Mov10) protein in HepG2 cells and investigate the role of Mov10 in the cell proliferation and invasion. Methods The eukaryotic expression plasmid pCMV-Mov10-GFP was transfected into the HepG2 cells mediated by lipofectamine and then selected with G418. The mRNA and protein expression of Mov10 was detected by qPCR and Western blotting
respectively. Immun
ofluorescence assay was used to analyze the location of Mov10. The proliferation and colony-formation ability was measured by MTS assay and plate colony-formation assay. Cell invasion assay was performed
in vitro
. Results Our established HepG2-Mov10 Cells stably expressed Mov10 protein. Endogenous and exogenous Mov10 protein was mostly distributed in the cytoplasm. HepG2-Mov10 cells had significantly higher proliferative rate and stronger ability of invasion than HepG2 cells (
P
<
0.05). Colony forming efficiency was (56.7±17.3)%
(38.3±12.1)%
(31.3±8.1)% respectively in HepG2-Mov10
HepG2-GFP and HepG2 cells
with the fromer significantly higher than the other 2 cell lines (
P
<
0.05). Conclusion A hepatoma stable cell line over-expressing Mov10 is successfully established. Stable overexpression of Mov10 protein obviously promotes cell proliferation
colony formation and invasion.
