摘要

采用单克隆抗体（单抗）和ＷＨＯ６７／９７标准血清建立ＥＬＩＳＡ法测定人血清免疫球蛋白Ｇ（ＩｇＧ）亚类浓度。结果表明：本法敏感（最低可测值为２．１ｎｇ／ｍｌ，可测范围为１００倍），精确（不同批测定平均变异系数≤８．０６％）。所用单抗特异性强，反应性好，ＩｇＧ亚类之和与常规单扩法所测总ＩｇＧ值密切相关（ｒ＝０．９６，Ｐ＜０．０１）．强调将单抗包被于经ＢＳＡ－戊二醛处理的微孔板上，能明显提高单抗吸附力和反应性；两种单抗混合包被可增加ＩｇＧ亚类同种异型覆盖面。

Abstract

ith monoclonal antibodies(McAbs )and 67/97 Standard sera

Serum concentrations of normal humanIgG Subclasses were measured by ELISA. The results showed that our method was sensitive( 2. 1ng/ml ofthe lowest detectable value

range 100x) and accurate(≤8. 06%of CV in various detectious).McAbs used inour measurement were excellent in the specifity and responsibility.Total values of IgGmeasured with ELISA were related to that with the single diffusion assayt(r=0. 96.P<0)01). It was emphasized that coating McAbs on the microtitre well pretreated with BSA-glutaraldehyde couldprovide the adherence and responsitivity of McAbs

and the coating with the two mixed McAbs could enlargethe covering field of igG subclass isotypes.
