摘要

为准确地在体内评价Ｗｉｓｔａｒ大鼠的胰岛素敏感性，我们建立了大鼠的正常血糖－高血浆胰岛素钳夹技术（简称钳夹技术）．并运用该技术初步探讨了年龄，体重、血浆胰岛素及血脂水平对胰岛素敏感性的影响。结果显示，我们建立的钳夹技术所测定的葡萄糖输注率（ＧＩＲ）、细胞外液变化量及其它指标均接近或优于文献报道。同时发现，年龄、体重大于Ⅰ组大鼠的Ⅱ组大鼠（Ｐ＜０．０１）．其ＧＩＲ、血浆高密度脂蛋白组分２水平显著低于Ⅰ组大鼠（Ｐ＜０．０１）．而血浆胰岛素及血浆甘油三酯水平显著高于Ⅰ组大鼠（Ｐ＜０．０１和０．０５）。本研究的结果提示：（１）我们建立的钳夹技术较准确、可靠（２）随着年龄、体重的增大，大鼠的胰岛素敏感性逐渐下降。

Abstract

o assess in vivo insuin sensitivity of Wistar rats

we developed the euglycemic - hyperinsulinemic clamp(E C)technique and investigated the effect of age

plasma insulin and plasma lipid level on insulin sensitivity with EC technique. The results showed that glucose infusion rate (GIR)

the expansion of extra - cellular fluid and other indices of the rats determined by EC technique were similar to those of the references. Moreover

in rata group Ⅱwho were older of heavier than rats group Ⅰ(P<0.01)

GIR

plasma high density lipoprotein subclass 2(HDL2 ) were lower than those in rats group Ⅰ(P< 0.01)

and plasma insulin. plasma triglycerides level were higher than those in rats group Ⅰ(P <0.05

respectively). The study suggests that the E. technique which we developed is relatively reliable and accurate. Besides . insulin sensitivity in Wistar rats declines with age and weight.
