摘要

目的:探讨青蒿酸体外作用对K562细胞增殖的影响及其机制。方法:采用MTT法检测青蒿酸对K562细胞增殖的影响

流式细胞术分析细胞周期分布情况

透射电子显微镜进行形态学观察。结果:青蒿酸作用K562细胞24h、48h

能显著抑制细胞增殖

随药物浓度增加

对细胞增殖的抑制强度也显著增高。经青蒿酸作用48h

处于G0/G1期K562细胞增加

且能诱导K562细胞发生凋亡。电镜可观察到细胞凋亡的形态学改变。结论:青蒿酸能抑制K562细胞增殖

这种抗肿瘤作用可能与抑制K562细胞进入细胞增殖周期和诱导细胞凋亡有关。

Abstract

Objective: To investigate the effects of artemisinic acid on proliferation of K562 cells and its mechanism in vitro.Methods: MTT assay was used to detect the effects of artemisinic acid on proliferation of K562 cells.The flow cytometry was used to analyze the distribution of cell cycle.Morphological changes of cells were observed by electron microscopy.Results: The proliferation of K562 cells was inhibited by intervention of artemisinic acid significantly after 24 hours and 48 hours

and the effects were in concentration-dependent manner.The cell number in G0/G1 increased and apoptosis was induced after the treatment with artemisinic acid for 48 hours

the morphological characteristic of apoptosis was observed by electron microscopy.Conclusion: Artemisinic acid can inhibit the proliferation of K562 cells

which may be-due to induction of apoptosis and prohibit on of tumor cells from entering cell cycle.
