In vitro study on the effect of bone marrow mesenchymal stem cells on the fatty acid metabolism of hypertrophic cardiomyocytes
Objectives:To inverstigate the effect of bone marrow mesenchymal stem cells(MSCs) on the fatty acid metabolism of hypertrophic cardiomyocytes and disclosure its mechanisms. Methods:Wistar neonate rat cardiomyocytes cultured in vitro were randomized into an angiotensin(Ang)-Ⅱstimulating group(n=5)
a MSCs treated Ang-Ⅱ stimulating group(Intervention group
n=5) and a normal control group(n=5).The activity of nuclear factor(NF)-κB and peroxisome proliferators-activated receptor(PPAR) α and γ were detected by immunofluorescence microscope
and intracellular free fatty acids(FFA) content by ultraviolet spectrophotometry.Results:Ang-Ⅱstimulated the NF-κB activation of neonate rat cardiomyocytes to strengthen
inhibited the activation of PPARα and PPARγ to weaken and increased the accumulation of FFA(P<0.01).For MSCs intervention group
the activation of NF-κB and the accumulation of FFA decreased significantly(P<0.01 and P<0.01) while the activation of PPARα and PPARγ were up-regulated compared with the Ang-Ⅱstimulating group(P<0.05 and P<0.01).Conclusion:MSCs could improve the fatty acid metabolism of hypertrophic cardiomyocytes
which is probably one of the mechanisms of the marrow mesenchymal stem cell transplantation improving the heart functions
and NF-κB and PPARs may be the signal passage of function.
