摘要

目的:探讨骨形态发生蛋白-2（Bone morphogenetic protein-2

BMP-2）、热化疗单独和联合应用对人结肠癌细胞SW480株增殖凋亡的影响并探讨其可能机制。方法:BMP-2（25、50、100、200、400μg/L）作用于SW480细胞株

MTT法检测其细胞活力。BMP-2、五氟尿嘧啶（5-Fluorouracil

5-FU）单独与联合作用于SW480细胞株

流式细胞（Flow cytometry

FCM）检测细胞凋亡率

Western blot检测人第10号染色体缺失的磷酸酶及张力蛋白同源的基因（Phosphatase and tensin homolog deleted on chromosometen

PTEN）蛋白的表达

逆转录聚合酶链式反应（RT-PCR）检测PTEN mRNA表达。结果:BMP-2可以抑制SW480细胞的增殖。与单独作用比较

BMP-2联合热化疗作用于SW480细胞株时

其凋亡率明显增高（P<0.05）。BMP-2能够提高PTEN蛋白的表达水平

但是不能提高PTEN mRNA的表达水平。结论:BMP-2可以提高热化疗的敏感性。BMP-2与热化疗联合作用有协同诱导SW480细胞株凋亡的作用

其机制可能与上调PTEN蛋白表达及阻止PTEN蛋白降解有关。

Abstract

Objective:Investigate effects of bonemorphogeneticprotein-2(BMP-2)

Thermo-chemotherapy and their combination on proliferation and apoptosis in human colon cancer cell(SW480) line

and to explore its probable mechanisms.Methods:SW480 cells were co-cultured with different concentrations of BMP-2(25

the apoptosis rate was detected by FCM.The protein level of PTEN was detected by Western blot.The mRNA expression of PTEN was detected by reverse transcription PCR.Results:BMP-2 can inhibit the proliferation of SW480 cells.The apoptosis rate of SW480 cell treated Thermo-chemotherapy+BMP-2 was significantly higher than that of BMP-2 treated and that of Thermo-chemotherapy treated cell(P<0.05).BMP-2 can improve the expression of PTEN protein

but can not improvethe expression of PTEN mRNA.Conclusion:BMP-2 can improve the sensitivity of Thermo-chemotherapy.BMP-2 combined with Thermo-chemotherapy can induce apoptosis of SW480 cell line

through its up-regulation of the protein level of PTEN and inhibition of PTEN protein degradation.
