摘要

目的分析口腔鳞状细胞癌组织中长链非编码RNA（lncRNA）肺腺癌转移相关转录因子1（MALAT1）的表达及其临床意义。方法收集口腔鳞癌组织标本81例和正常口腔黏膜组织20例，分别设为口腔鳞癌组和健康对照组。采用RTPCR方法检测2组组织中lncRNA MALAT1表达情况，统计分析lncRNA MALAT1表达与病理资料特征的关系，KaplanMeier分析lncRNA MALAT1表达和生存预后间的关系。结果与健康对照组相比，lncRNA MALAT1在口腔鳞癌组中表达明显增高，差异有统计学意义（P<0.05）；口腔癌组织中lncRNA MALAT1的相对表达量在不同肿瘤分期、肿瘤分化程度和有无颈部淋巴结转移间比较，差异均有统计学意义（P＜0.05）；而在不同年龄、性别、肿瘤大小及肿瘤类型间比较，差异均无统计学意义（P>0.05）；lncRNA MALAT1高表达组和低表达组患者的3年总体生存率(OS)分别为45.9%（17/37）、65.9%（29/44），KaplanMeier生存分析（log-rank检验）表明MALAT1高表达组患者3年OS显著低于MALAT1低表达组患者，差异有统计学意义(P<0.05)。结论口腔鳞癌组织中lncRNA MALAT1表达增高，其表达水平与口腔鳞癌患者的不良预后有关，在口腔鳞癌组织中检测其表达可能有助于肿瘤的诊断及作为药物治疗的靶点。

Abstract

Objective To analyze the expression of long noncoding RNA(lncRNA) metastasisassociated lung adenocarcinoma transcript 1(MALAT1) in oral squamous cell carcinoma and its clinical significance.Methods A total of 81 cases of oral squamous cell carcinoma tissue specimens and 20 cases of normal oral mucosa tissue specimens were collected and divided into the oral squamous cell carcinoma group and the healthy control group.The expression of lncRNA MALAT1 in tissues of the two groups was detected by RTPCR

and the relationship between the expression of lncRNA MALAT1 and pathological data were analyzed.KaplanMeier was used to analyze the relationship between the expression of lncRNA MALAT1 and survival prognosis.Results Compared with the healthy control group

the expression of lncRNA MALAT1 in oral squamous cell carcinoma group was significantly increased(P<0.05).There were significant difference in the relative expression of lncRNA MALAT1 at different stages of oral cancer

degree of differentiation and cervical lymph node metastasis(P<0.05)

but there was no significant differences in age

sex

size and type of tumor(P>0.05).The 3year overall survival rate(OS) of the high and low expression groups of lncRNA MALAT1 was 45.9%(17/37) and 65.9% (29/44)

respectively.KaplanMeier survival analysis(logrank test) showed that OS in high MALAT1 expression group was significantly lower than that in low MALAT1 expression group in 3 years (P<0.05).Conclusion The expression of lncRNA MALAT1 is increased in oral squamous cell carcinoma.The expression level of lncRNA MALAT1 is related to the poor prognosis of oral squamous cell carcinoma patients.Detection of the expression of lncRNA MALAT1 in oral squamous cell carcinoma tissues may be helpful for the diagnosis of tumors

and it may be a target for drug therapy.
