Mechanism of moist exposed burn ointment promoting wound repair in diabetic mice based on the GRP78/PERK/CHOP signaling pathway mediated by endoplasmic reticulum stress
To investigate the effect of moist exposed burn ointment(MEBO) on the expression of the glucose-regulated protein 78(GRP78)/protein kinase R-like endoplasmic reticulum kinase(PERK)/CCAAT/enhancer-binding protein homologous protein(CHOP) signaling pathway in diabetic wounds.
Methods
2
A total of 27 spontaneous diabetic db/db male mice were randomly divided into model group,MEBO group, and recombinant bovine basic fibroblast growth factor(rb-bFGF) gel group, and then 9 heterozygous db/m male mice of the same strain without disease were selected as control group. The mice in the MEBO group and the rb-bFGF group were given intervention with the external application of MEBO and rb-bFGF,respectively,and those in the control group and the model group were given Vaseline for intervention. The mice were observed daily, and wound healing was calculated on days 3,6,and 12 of intervention. Western blot and immunohistochemistry were used to measure the protein expression levels of GRP78,PERK and CHOP in the wound tissue of each group on days 3,6,and 12.
Results
2
On day 3 of wound intervention, the control group had a significantly higher wound healing rate than the model group(
P
<
0.01),and on days 6 and 12 of wound intervention, the control group, the MEBO group, and the rb-bFGF group had a significantly higher wound healing rate than the model group(
P
<
0.01). On day 3 of wound intervention, there were no significant differences in the protein expression levels of GRP78,PERK,and CHOP in wound tissue between groups(
P
>
0.05),and on days 6 and 12 of wound intervention, the model group had significantly higher protein expression levels of GRP78,PERK,and CHOP in wound tissue than the control group, and the MEBO group and the rb-bFGF group had significantly lower protein expression levels of GRP78,PERK,and CHOP than the model group(
P
<
0.05).
Conclusion
2
MEBO may inhibit excessive endoplasmic reticulum stress and promote diabetic wound healing by inhibiting the expression of the GRP78/PERK/CHOP signaling pathway.
