Effect of urine-derived stem cell transplantation on recovery of neurogenic bladder in rats after spinal cord injury
Objective To observe the function and histological changes of neurogenic bladder
and investigate the effect of urine-derived stem cell transplantation on the neurogenic bladder in rats after spinal cord injury (SCI). Methods Forty-five Sprague-Dawley (SD) rats were randomly divided into normal group
SCI-induced group
and SCI-induced and urine-derived stem cell transplantation group (n=15 for each group). On the 7th day after induction of SCI
the rats in the stem cell transplantation group were injected with urine-derived stem cells. After 28 d
urodynamic test
in vitro detrusor smooth muscle strips and bladder wet weight were used to measure the function of bladder. Western blotting was employed to detect the expression of P2Y4 in each group. TUNEL assay was performed to test the apoptosis ratio of bladder cells. Results The SCI-induced and urine-derived stem cell transplantation group was better in bladder wet weight (0.31±0.01 g)
in vitro detrusor smooth muscle strips (contraction intermittence 16.47±2.29 s)
and bladder cell apoptosis ratio [(28.22±11.08)%] compared with the SCI-induced group [0.66±0.07 g
2.30±0.26
59.00±5.68 s
19.80±3.77 s
(59.80±14.18)%
P0.05]. Western blot analysis showed that the expression of P2Y4 was reduced in the bladder of the SCI-induced and urine-derived stem cell transplantation group. Conclusion Urine-derived stem cells can promote recovery of neurogenic bladder in rats after SCI.
