Small intestinal submucosa cryogel as scaffold for cell transplantation
Objective To prepare a new scaffold with shape memory properties for cell transplantation by using porcine small intestinal submucosa (SIS). Methods The acellular SIS powder was dissolved
mixed with different concentrations of 1-ethyl-3-(3-dimethyllaminopropyldvb)carbodiimide hydrochloride (EDC) with a self-made mixing device
and crosslinked in the frozen condition. The elastic deformation and shape memory capability were detected. Scanning electron microscopy was used to perform morphological observation and detect the pore size
and the water absorption ability
biodegradation behavior and mechanical properties were detected. The mice fibroblasts were used for cell adhesion and proliferation study. Results SIS cryogel could be compressed over 85% and injected through a needle without any damage. The cryogel had porous and interconnected pore structure with average pore size of 106.63±28.90 μm. There was no significant difference in water absorption ability and cytotoxicity at different concentrations of EDC. But the degradation became slow and mechanical strength was enhanced when the EDC concentration increased. Cells grew linearly and spindly on the SIS cryogel
and stretched out plenty of pseudopodiums. Some sections were integrated into film and stuck to the surface and pore walls. Conclusion Injectable SIS cryogel with elastic deformation and shape memory capability can be used as cell transplantation scaffold because of good biocompatibility and physical properties.
