摘要

目的观察利用微流控免疫磁珠分选仪富集外周血循环肿瘤细胞（circulating tumor cells，CTCs）的效果。方法采用IsoFlux系统（微流体免疫磁珠分选系统），对16例肺癌患者（TMN分期法，Ⅰ～Ⅳ各4例患者）外周血中循环肿瘤细胞进行分离富集，并通过免疫荧光的方法标记CTCs的标记蛋白，对分选出的CTCs细胞进行了鉴定。结果 16位肺癌患者血样中10位患者的血样中均发现了Cytokeratine阳性和CD45阴性的CTCs，Ⅰ～Ⅳ期各4例患者的血样中检出CTCs细胞检出率分别为50%、50%、75%和100%。结论利用微流控免疫磁珠分选仪可以有效地富集外周血中稀有的循环肿瘤细胞，具有较高的灵敏性及特异性。

Abstract

Objective To establish a method for isolation and enrichment of circulating tumor cells (CTCs) from peripheral blood utilizing Isoflux system (microfluidic immunomagnetic bead sorter). Methods IsoFlux system was used to separate and enrich CTCs in peripheral blood of 16 patients with lung cancer (clinical stage Ⅰ~Ⅳ

and the marker protein of CTCs was marked and then identified by immunofluorescence assay. Results There were 11 patients with cytokeratine-positive and CD45-negative CTCs

and the detection rates of CTCs in clinical stages Ⅰ~Ⅳ were 50%

respectively. Conclusion CTCs can be simply isolated and enriched from peripheral blood using the IsoFlux system at high specificity and sensitivity.
