Alteration of circulating eosinophila microparticles in asthmatic patients before and after treatment
Objective To explore the changes of circulating eosinophila microparticles in the patients with asthma
and investigate the relationship of the changes with treatment efficacy. Methods Fifty-seven outpatients of our department from April to September 2015 who met the inclusion criteria were enrolled in this study. They were 32 patients of the asthma group and 25 of the normal control group. Their peripheral venous blood samples were collected
centrifuged and then marked with corresponding antibodies. Then the numbers of eosinophila microparticles in every 100 μL supernate was determined by flow cytometry. After 3 months of regular anti-asthmatic treatment (inhaled corticosteroids + long-acting beta 2 agonists)
the equivalent volume of venous blood was harvested from the subjects of asthma group and then extracted for the detection of the numbers of eosinophila microparticles. Results The circulating level of eosinophila microparticles was significantly higher in the asthma group before treatment than that in control group and the asthma group after treatment (F=5.532
P=0.038; Z=-4.937
P0.01)
and there was no statistical difference between the asthma group after treatment and the control group (F=3.118
P=0.102). The number of microparticles has no significant difference between the allergic asthma subgroup and the non-allergic asthma subgroup (t=0.421
P=0.677; t=0.006
P=0.952)
but statistical difference was seen in the number between the well-controlled asthma group and un-controlled group after treatment (t=-2.086
P=0.046)
and Δ eosinophila microparticles represented linear correlations with the changes of forced expiratory volume in 1 second (FEV1)
the percentage of FEV1 (FEV1%)
eosinophil count (ΔEos count)
faction exhaled nitric oxide (FeNO) and athma control test (ACT) (r=-0.558
P0.01; r=-0.453
P=0.005; r=0.625
P0.01; r=0.313
P=0.041; r=-0.399
P=0.012). Conclusion The number of circulating eosinophila microparticles is increased in asthma patients
and decreased after treatment. So
dynamical monitoring its level may contribute to the assessment of asthma control.
