Radiosensitizing effect of 4-HPR on human cholangiocarcinoma cell line QBC939
Objective To explore the radiosensitizing effect of fenretinide (4-HPR) on human cholangiocarcinoma cell line QBC939. Methods MTT assay was used to determine the half maximal inhibitory concentration (IC
50
) of 4-HPR to QBC939 cells. Radiosensitization of 4-HPR was measured by clone formation assay
and survival curve was described. Cell apoptosis was tested by flow cytometry. The apoptosis-assoc
iated proteins were analyzed by Western blotting. Results 4-HPR inhibited the proliferation of QBC939 cells in a concentration-dependent manner. Compared with the single radiotherapy group
the 4-HPR+radiotherapy group showed lower cell survival rate. The mean lethal radiation doses (D
0
) of the single radiotherapy group and 4-HPR+radiotherapy group were 2.72 Gy and 1.93 Gy
respectively
and the prospective domain doses (D
q
) were 2.06 Gy and 0.94 Gy
respectively. The gain ratio of radiotherapy (SER) of 4-HPR was 1.41. After 4-HPR treatment
the cell apoptosis was increased
showing low expression of bcl-2 and high expression of caspase-3. Conclusion 4-HPR inhibits the growth of QBC939 cells and enhances the radiosensitivity of QBC939 cells in vitro
