Correlation of glutathione S-transferase polymorphisms with sperm mitochondrial membrane potential in idiopathic male infertility
Objective To investigate the correlation of glutathione S-transferase M1
T1 and P1 gene (GSTM1
GSTT1 and GSTP1) polymorphisms with sperm mitochondrial membrane potential (MMP) in idiopathic male infertility. Methods A total of 246 patients with idiopathic male infertile admitted in the First Affiliated Hospital of Medical College
Xi’an Jiaotong University from September 2008 to June 2012 were enrdled in this study. Polymerase chain reaction (PCR) and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) were used to identify the GSTM1
GSTT1 and GSTP1 genotypes in the sperm . Then flow cytometry using 5
5′
6
6′-tetrachloro-1
1′
3
3′-tetraethylbenzimidazolylcarbocyanine (JC-1) as staining agent was employed to detect the sperm MMP. Results The percentage of normal sperm MMP was significantly lower in the GSTM1 and GSTT1 deletion groups than the wild type group in idiopathic male infertile patients (P0.01)
and the 2 type of gene detection had synergistic effect. However
there was no significant difference in the percentage of normal sperm MMP between the GSTP1 mutation type group and the wild type group (P=0.085). Conclusion GSTM1 and GSTT1 deletions are risk factors for the sperm MMP in the idiopathic male infertile patients.
